How would one inoculate 1/2 pint jars that have filter discs with a syringe?

By Brettiejams (Brettiejams) on Saturday, September 29, 2001 - 04:12 am:

In a Glove-Box, in front of a Flow Hood, or using the Oven Rack Method.

You need to open the top of the jar to get the innoculant in(do not pierce the filter disk with the syringe)

I just inject right through the discs. The discs are cheap enough to replace when i use 100 of them or you can fill in the hole with either tape or silicone sealant. I just cap the hole with electrical tape. 100% colonization time on my last 5 jars of thailand strain from a spore syringe and brown rice + brown rice flour... 14 days (no shit)

If you're gonna go and do that then you should just use Poly-Fil.... MUCH cheaper and less prone to Contamination. Ive gotten contams growing in those discs if I dont keep them sprayed down with lysol or the equivelent.

By littlebro (Littlebro) on Saturday, September 29, 2001 - 08:33 am:

those discs can get infected with mold/bacteria/stuff and contaminate rye grain jars, even after pcing. . haven't tried them with half pints, they're probably money that way.

I was thinking of a purchase from Fungi Perfecti and would appreciate a little comment from folks. Are the synthetic filter discs as good or better than verm as a barrier for contams? What size are half pints? Are the autoclavable bags with patches good for casing material to sterilize in a pc?Thank you yall

By Fishy1 (Fishy1) on Friday, November 30, 2001 - 03:53 am:

I wont use the filters...but if contam probs exist, feel free.
The bags will work well for casing mix. I used heavyduty foil, and wrapped my peat 50/50 in it like a loaf. Worked GREAT. And cheaper, too.
IMO, I would save the effort and $...but I am fishy1
--some of the bags that soil, vermic, etc...come in are PC-able.

i wouldn't waste expensive filter patch bags to sterilize casing materials. use oven bags or non filter patch autoclavable bags. you can replace the verm layer with Polyfill. filter disc's work great, but if you're using the pftek, how are you going to inoculate through the disc? you don't want to poke holes in it.

poly-pf

if you look just below the lid you can see the white polyfil that is replacing the verm layer.

poly-pf2

here's a close up of an inoculation hole. notice it isn't taped. i've never taped inoc. holes with the verm layer so i figured why do it w/ a polyfil layer. not taping holes has never caused any problems at all. put foil over the lids before sterilizing.

again, unfortunately Polyfil is not very compatible w/ blunted syringes. although i did do a few jars with one blunted syringe and it did work 100% but i had to shoot the solution partially through the poly. not so good as far as getting the solution down the jar very far. nonblunted syringes worked like a dream.

what about shooting up Karo Tek jars? any tips? iv read the archives but i havnt heard to many people talking about this. im scared to try because the polyfill. but only thing i got is blunt syringe tips :\ anyone have any good tips on the Karo tek jars, do you keep foil on even after inocculation, im scared about contams.

By relic (Relic) on Friday, November 30, 2001 - 06:09 am:

if you have blunted tips, you can use a separate inoculation hole in the lid covered with tape for the honey or karo tek(although nan mentions no vent in the karo tek). you can also do this with qts. of grain with polyfil vents if you have blunted syringes.

Fishy,

Do you put the casing material in foil then in the oven or pc?I would be using the fifty, fity.Should I bake it for how ong and at what temp? Thanks

By Fishy1 (Fishy1) on Friday, November 30, 2001 - 08:46 pm:

I put the moist foil loaf in the PC for 1 hr or so.
You could bake it. Maybe 350 for an hour? I dunno....
Works great! I like coarse vermiculite in mine.
I have an experiment in the cooker right now-straw, worm castings, vermic, cottonseed meal, etc...in a large gusseted plastic coffee bag. We'll see how that works! If I am not back in an hour, send for help!!!

Just a question regarding filter disks, and their uses. They seem like almost a perfect solution for anyone concerned with gas exchange problems for both Honey Tek and PF Tek, so my question is, is it actually possible to use just filter disks as lids for both of those Teks? If they are, then what is the main reason that no one uses them? Is it just a cost factor, or is there something else?

they could be used as you suggest, but their primary drawbacks are 1] they allow water vapor to escape, potentially drying out pf cakes prematurely. 2] if they get wet with honey/dextrose solution, contams can colonize the filter itself and enter the jars.
3] they have no holes, so you must work in a glovebox when inoculating.
i still have dozens of unused filter disks, unused since i switched to polyfil, which is cheaper, more adaptable, and allows needles to easily penetrate.

Personally Mr. Hicks, I think for the most part they are a waste of money. Notice that the guy who sells them, pushes them the MOST in the newbie market.

And yes, you can use filter disks for whatever you want. But IMHO if you are at least halfway sterile, you don't need them.

And, go to a medical supply warehouse, they have them in every town, and pick up micron/particulate
face mask filters for WAY cheaper than he sells them, not only are they HOSPITAL quality, but you get more product.

Just my opinion, prolly worth about the pixels it is showing on. A guru will probably come in any minute to help you ( quote has been following me around all morning ;0) )
